摘要
目的:研究芸香科植物两面针HPLC指纹图谱与抗宫颈癌活性之间的关系。方法:对33批两面针药材进行HPLC分析,以人宫颈癌细胞株HeLa为药效模型,用MTT法测定各批次两面针药材的抗肿瘤活性,再通过多元线性回归法,建立两面针抗宫颈癌谱-效关系方程。结果:建立了两面针抗宫颈癌谱效关系方程,利用该方程对10批两面针药材进行抗宫颈癌活性预测,预测值与实际值的偏差率不超过10%,表明谱-效关系可用于预测两面针的抗宫颈癌作用。结论:两面针抗宫颈癌谱-效关系方程可反映抗肿瘤作用是两面针药材中多种成分协调、互补或制约相互作用的结果,谱-效关系研究为有效地控制两面针质量搭建新的平台。
Objective: To study the relationship between HPLC fingerprint chromatogram and inhibitory effect on anticancer activity of Zanthoxylum nitidum.Method: Zanthoxylum nitidum fingerprint was established,Tumor cell lines HeLa was chosen as efficacy model,the anticancer activity of 33 sources of Zanthoxylum nitidum extracts were determined using MTT method.Multiple linear regression was applied to construct a multivariate regression model.Results: The chromatogram-effect relationships on HeLa was established,10 Zanthoxylum nitidum samples were chosen for verification,the deviation of predictive value and true value was not exceed 10%.The result indicated that the multiple linear regression could predict the anticancer activity to HeLa from the fast chromatograms.Conclusion: The chromatogram-effect relationships on HeLa has good predictive and describing abilities to evaluate the quality and therapeutic activity of Zanthoxylum itidum.It is more reasonable to evaluate the quality of traditional Chinese medicine by the bioactivity-fingerprint.
