摘要
目的:研究超支化聚醚酯-二氢卟酚e6纳米光敏剂[hyperbranched poly(ether-ester)-chlorin e6 nano-photosensitizer,HPEE-ce6]的构建过程与表征,及其对人舌鳞癌CAL-27细胞(Tongue cancer CAL-27 cells)体外光动力疗法(Photodynamic therapy,PDT)的杀伤作用。方法:通过一步合成法构建HPEE-ce6纳米光敏剂,用紫外可见吸收光谱和透射电镜观察其表征,通过透射电镜观察CAL-27细胞的形态学改变。结果:HPEE和ce6共价结合,生成直径为50 nm左右的纳米粒子,其治疗区吸收峰比ce6红移了12 nm;经PDT处理后,形态学研究表明,CAL-27细胞发生凋亡或特征性肿胀样坏死。结论:HPEE-ce6可以通过一步法(HPEE和ce6共价交联)合成,并对人舌癌CAL-27细胞具有良好的光动力杀伤作用。
PURPOSE: To construct and study the characteristics of hyperbranched poly(ether-ester)-chlorin e6 nano-photosensitizer(HPEE-ce6);and the cytocidal effect of HPEE-ce6 on tongue cancer CAL-27 cells.METHODS: HPEE-ce6 nanoparticles were prepared by reaction of hyperbranched poly(ether-ester)(HPEE) and chlorin e6(ce6) by one-step method,and characterized by ultraviolet-visible absorption spectroscopy(UV-Vis) and transmission electron microscopy(TEM).The morphological changes of the CAL-27 cells were observed by TEM.RESULTS: The nanoparticles with a diameter of 50nm were synthesized by covalent binding between HPEE and ce6,and the absorption peak of HPEE-ce6 nanoparticles was red shifted 12 nm compared with ce6.CAL-27 cell apoptosis or characteristic oncosis was observed after CAL-27 cells were treated by PDT.CONCLUSION: The results indicate that HPEE-ce6 nanoparticles could be synthetized by one-step method;the cytocidal effect of HPEE-ce6 was good with photodynamic therapy on CAL-27 cells in vitro.
