摘要
观察不同剂量丙酸睾丸酮对MCF-7荷瘤裸鼠肿瘤生长的影响,并探讨其可能机制和临床意义。构建MCF-7荷瘤裸鼠,再用不同浓度的丙酸睾丸酮(50mg/kg,400mg/kg)及化疗药物(表阿霉素)进行干预,比较各组实验鼠肿瘤的生长情况,并用微粒子发光法检测裸鼠血清睾丸酮的含量,用流式细胞技术检测肿瘤中雄激素受体的表达。发现给予低剂量丙酸睾丸酮(50mg/kg)组的裸鼠肿瘤体积明显增大且明显高于对照组及化疗组(P=0.047,0.034),给予高剂量丙酸睾丸酮(400mg/kg)组裸鼠的肿瘤生长不一,三只肿瘤消褪,二只肿瘤生长。用药结束后400mg/kg丙酸睾丸酮组实验鼠血清睾丸酮水平明显升高,与50mg/kg组、化疗组及对照组相比差异有统计学意义(P=0.019,0.000,0.000)。各组肿瘤中雄激素受体表达差异无统计学意义(P=0.177)。低剂量丙酸睾丸酮可刺激MCF-7乳腺癌裸鼠肿瘤的生长,高剂量丙酸睾丸酮可能抑制MCF-7乳腺癌荷瘤裸鼠肿瘤的生长,但其作用并不明确,具体机制仍待进一步研究。雄激素对乳腺癌的治疗还需谨慎对待。
To investigate the effect of testosterone propionate (TP) at different dose on the growth of MCF--7 xenograft tumor in nude mice and to explore it may--be mechanism and clinical significance. The transplanted MCF--7 tumor in nude mice was established. Then the mice were treated by testosterone propionate at different dose (50mg/kg, 400mg/kg) and chemotherapy medicine. Tumor growth status was analyzed. Microparticie cheniiuminescance immunoassay was per- formed to measure the level of serum testosterone of nude mice. FCM was performed to examine the expression of AR in the transplanted tumors. The volume of transplanted tumor in lower dose TP group increased significantly compared with control group and chemotherapy group (P=0. 047, P= 0. 034, respectively). The growth of transplanted tumors in high dose TP group was complex. 3 transplanted tumors were gradually reduced, but 2 transplanted tumors became bigger ob- viously. The highest testosterone level was found in high dose TP (400mg/kg) group, which was significantly higher than the other three groups(P= 0. 000, P= 0. 000, P = 0. 019, respectively). No significance of AR expression was found among those groups (P=0. 177). Lower dose TP can stimulate the MCF--7 transplanted tumor. Higher dose TP may be inhibit the growth of MCF--7 transplanted tumor, but its effect is unclear and needs further approach.
