参麦总皂苷对人脐静脉内皮细胞血管细胞黏附分子-1表达影响的研究

摘要 目的:研究参麦总皂苷对人脐静脉内皮细胞(HUVEC)血管细胞黏附分子-1(VCMA-1)表达的影响,探讨参麦抗肿瘤作用的可能机理。方法:制备参麦总皂苷,以炎症因子TNF-α为诱导物,用一系列浓度的参麦总皂苷进行干预处理HUVEC细胞,采用荧光定量PCR方法和流式细胞仪检测VCMA-1基因的转录和表达情况。结果:TNF-α可剂量依赖性诱导HUVEC细胞表达VCMA-1,其中20ng/ml为合适诱导浓度;参麦总皂苷可以从mRNA水平和蛋白质水平显著抑制TNF-α诱导的VCMA-1表达,并且与剂量呈正相关,但对正常HUVEC细胞的VCMA-1表达没有明显影响。结论:参麦总皂苷对TNF-α诱导的HUVEC细胞VCMA-1表达有抑制作用,提示其具有抗肿瘤转移作用。 Objective: To study the effect of total saponins in shenmai on the expression of vascular cell adhesion molecule-1 (VCMA-1) in human umbilical vein endothelial cell(HUVEC) and discuss the possible molecular mechanism of anti-tumor effect of shenmai. Methods: Preparing total saponins from shenmai injection, adopting TNF-α to induce the the expression of vascular cell adhesion molecule-1 (VCMA-1) in human umbilical vein endothelial cell and intervened with a series of concentration of total saponins from shenmai, the gene transcription and protein expression of vascular cell adhesion molecule-1 was determined by fluorescence quantitative PCR and flow cytometry. Results: TNF-α could induce HUVEC to express VCMA-1 in the shape of dose-effect relationship, 20ng/ml was the adequate induce concentration; total saponins in shenmai could inhibit the gene transcription and protein expression of vascular cell adhesion molecule-1 in HUVEC induced by TNF-α, the effect present positive correlation, but there was no effect on VCMA-1 expression in normal HUVEC. Conclusion: Total saponins in shenmai can inhibit the expression of vascular cell adhesion molecule-1 in human umbilical vein endothelial cell induced by TNF-α, which suggests that total saponins in shenmai can control the cancerometastasis.
