摘要
目的检测口腔鳞癌及癌前病变中DJ-1和PTEN的表达并分析其临床意义。方法免疫组织化学方法检测46例口腔鳞癌,16例口腔扁平苔藓,19例白斑和10例正常口腔黏膜标本中DJ-1、PTEN两种蛋白的分布,分析各个指标和病理参数的关系。结果口腔鳞癌组织中DJ-1增高,阳性率为89%(41/46),与正常口腔黏膜、口腔白斑和口腔扁平苔藓相比,差异有统计学意义(P=0.000);PTEN在口腔鳞癌组织中降低,阳性率为57%(26/46),与正常口腔黏膜、白斑和口腔扁平苔藓对比,差异有统计学意义(P=0.000)。经Spearman等级相关检验,DJ-1和PTEN之间的相关系数r为-0.67,P<0.01,差异具有统计学意义,表明DJ-1和PTEN之间存在负相关。结论 DJ-1、PTEN在口腔鳞癌发生过程中均起着一定的作用;DJ-1可能通过负性调控PTEN而导致口腔肿瘤的发生。
Objective To detect DJ-1 and PTEN protein in oral squamous cell carcinoma(OSCC) and precancerous lesions.Methods Immunohistochemical staining(SP method) was conducted to detect DJ-1 and PTEN in the biopsy mucosa tissues from 46 cases of OSCC,16 cases of oral lichen planus(OLP),19 cases of oral leukoplakia(OLK) and 10 cases of normal mucosa.Results The positive rate of DJ-1 in 46 cases of OSCC(89%) was significantly higher than that in other non-malignant groups,which included OLK,OLP and normal mucosa(P0.05),while the positive rate of PTEN in OSCC(57%) was significantly lower than that in other non-malignant groups(P0.05).A negative correlation was found between the expression of DJ-1 and PTEN(r=-0.67,P0.01).Conclusion Our data suggested that DJ-1 and PTEN may play an important role in the pathogenesis of OSCC,and DJ-1 may exert its function by negative regulating the expression of PTEN.
