摘要 目的:比较玻璃离子粘固剂(glass ionomer cement,GIC)、无机三氧化复合物(mineral trioxide aggregate,MTA)对MG-63细胞增殖和分化功能的影响。方法:配制GIC、MTA两种材料的浸提液,浓度分别为10g/L、20g/L、40g/L,使用不同浸提液对MG-63细胞进行培养,对照组为DMEM培养基。72h后,分别测定不同浓度组细胞的增殖功能和碱性磷酸酶活性。结果:细胞增殖功能测定结果为MTA各浓度组的A值均较对照组高,并且随着浸提浓度的提高,A值增大,差别有统计学意义(P<0.05);GIC各浓度组的A值均较对照组低,随着浸提浓度的提高,A值减小,差别有统计学意义(P<0.05)。细胞碱性磷酸酶活性测定结果为MTA组A值均较对照组高,与对照组进行单因素方差分析,其中20g/L、40g/L浓度组差别有统计学意义(P<0.05);GIC各浓度组A值均比对照组低,其中40g/L浓度组与对照组进行进行单因素方差分析,差别有统计学意义(P<0.05)。结论:MTA与GIC相比能促进细胞的增殖与分化。 Objective: To compare the effects of mineral trioxide aggregate(MTA),glass ionomer cement(GIC) on the proliferation and differentiation of MG-63 cells.Methods: The leaching liquor of MTA and GIC of three different concentrations(40mg/ml,20mg/ml,10mg/ml) were prepared.Then MG-63 cells were incubated in condition medium(DMEM)containing various concentrations of the leaching liquor of MTA and GIC,while cells remained in condition medium(DMEM) were served as control.Over 72 hour-period,the proliferation and differentiation of MG-63 cells were evaluated.Results: The optical density(OD) values of each subgroup of MTA were all higher than control group.There was statistically significant difference between three subgroups of MTA and control group(P0.05).The OD values of each subgroups of GIC were all lower than control group,There was statistically significant difference in three subgroups of GIC and control group(P0.05).As to ALP activity,the OD values of each subgroup of MTA were all higher than the control group,statistically significant differences were found within the control group,20mg/ml and 40mg/ml subgroup(P0.05).The OD values of each subgroups of GIC were all lower than control group,statistically significant difference was found between 40mg/ml subgroup and control group(P0.05).Conclusion: MTA can stimulate the proliferation and differentiation of MG-63 cells.
