OsWRKY79基因克隆和植物表达载体的构建

Cloning of OsWRKY79 gene and construction of its plant expression vector

摘要 WRKY是植物体中重要的转录因子,克隆水稻OsWRKY79基因将为其在植物体内的定位和功能研究奠定基础。根据GenBank公布的OsWRKY79基因序列设计特异引物,以RT-PCR法从水稻幼苗中克隆OsWRKY79基因,将其亚克隆至含有GFP基因的pBinGFP表达载体中,经菌落PCR和酶切鉴定,成功构建了重组质粒pBinGFP-OsWRKY79。 WRKY proteins are important transcription factors in many plant.Cloning OsWRKY79 gene isolated from rice was conduct to identify the subcellular location of OsWRKY79 and function.A pair of specific primers designed according to the sequence of OsWRKY79 gene in GenBank,and the complete cDNA sequence was obtained by RT-PCR using the total RNA of rice seedlings as the template.The gene OsWRKY79 was inserted into the expression vector pBinCFP.The recombinant plasmid containing OsWRKY79 gene was identified by PCR amplification and digestion with restriction endonucleases.
