微囊化异种嗅鞘细胞移植联合中药修复大鼠脊髓损伤

摘要 目的:旨在探讨基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)在异种嗅鞘细胞移植联合中药干预下脊髓损伤中的作用。方法:将188只SD大鼠随机分为正常对照组、单纯损伤组、异种嗅鞘细胞移植组、异种嗅鞘细胞移植联合中药治疗组,损伤组又设1、3、7、14、28 d 5个时相点,免疫组织化学技术检测MMP-9的表达,同时电镜观察血脊髓屏障的变化。结果:脊髓损伤后血脊髓屏障的超微结构不同程度损害、MMP-9表达上调,经干预治疗后出现一定程度的恢复,MMP-9表达下调。结论:脊髓损伤后MMP-9表达水平增加与伤后血脊髓屏障损害的发生、发展密切相关,MMP-9在脊髓损伤后血脊髓屏障损害的病理生理过程中起重要作用。异种嗅鞘细胞移植联合中药对脊髓损伤有较好的修复作用。 Objective: To explore the function of(matrix metalloproteinase-9,MMP-9) in heterotransplantation of olfactory ensheathing cells combined with China herb intervention after spinal cord injury.Methods: Healthy 188 SD rats were divided into three groups as following: normal,injury control,transplantation of olfactory ensheathing cells,transplantation of olfactory ensheathing cells combined with China herb.Posttraumatic tissues were achieved at 1,3,7,14 and 28 d after spinal cord injury.The expression of MMP-9 with immunohistochemistry were measured in three groups respectively.Electron microscope was used to observe the changes in blood-spinal cord barrier.Results: Blood-spinal cord barrier was damaged in some extent recoveried;the expression of MMP-9 was obviously upregulated after spinal cord injury.The ultrastructure of blood-spinal barrier was partly recoveried and the expression of MMP-9 was decreased after intervention.Conclusion: Increase of the expressional levels of MMP-9 has a closely correlation with production and development of blood-spinal cord barrier damage after cervical spinal cord trauma.MMP-9 plays important roles in the pathophysiological process of blood-spinal cord barrier damage after cervical spinal cord trauma.Heterotransplantation of olfactory ensheating cells combined with China herb is effective in the repair of spinal cord injury.
