HIF-1α,VEGF-C在胃腺癌中表达及与淋巴结转移的关系

摘要 目的:研究缺氧诱导因子-1α、血管内皮生长因子-C在胃腺癌组织中的表达及与淋巴结转移的关系。方法:用免疫组化染色(S-P)法检测44例胃腺癌组织,20例胃正常组织中HIF-1α、VEGF-C的表达。结果:HIF-1α在胃腺癌组织标本中阳性率为59.01%(26/44),正常胃粘膜组织标本中表达阳性率25.00%(5/20),两组差异显著(P<0.05);VEGF-C在胃腺癌组织标本中阳性表达率为52.27%(23/44),正常胃粘膜组织标本中表达阳性率20.00%(4/20),两组差异显著(P<0.05);HIF-1α,VEGF-C在低分化腺癌中阳性率最高,但与癌组织分化程度间差异无显著意义(P>0.05),而在T1、2,T3、4表达差异显著(P<0.05),在淋巴结转移组与无淋巴结转移组表达差异显著(P<0.05);两者在年龄、性别、肿瘤部位方面无显著差异;在HIF-1α、VEGF-C共同表达组,淋巴结转移率显著高于非共同表达组;HIF-1α、VEGF-C在胃腺癌组织表达成正相关(rs=0.85,P<0.05)。结论:HIF-1α、VEGF-C在胃腺癌组织中表达上调,其阳性表达可能在胃腺癌发生、发展和淋巴结转移过程中起重要作用;胃腺癌组织中HIF-1α和VEFG-C蛋白阳性表达成正相关;HIF-1α和VEFG-C联合检测对预测胃腺癌淋巴结转移,可能有一定的价值。 Objective:The present studies aimed at studying the relationship between HIF-1α and VEGF-C expression and lymph node metastasis in gastric cancer tissues.Methods:Streptavidin-peroxidase(S-P) method was used for the immunohistochemical detection of HIF-1α and VEGF-C protein in paraffin embedded archival samples from 44 patients with gastric cancer and 20 normal gastric tissues.Results:Of the 44 gastric cancer 26 cases(59.1%) showed HIF-1α immunoreactivity in the cytoplasm of cancer cells.And 23 cases(52.27%) showed VEGF-C positive in cytoplasm and they were significantly higher than those in normal tissues(P0.05).Of the 44 gastric cancers 29 cases were lymph node metastasis;There were 21 cases that showed HIF-1α and 19 cases showed VEGF-C and they were significantly higher than those in no lymph node metastasis(P0.05);The expression of HIF-1α protein had significant positive correlation with the expression of VEGF-C protein in gastric cancers(rs=0.85,P0.05).Conclusions:The up regulated expression of HIF-1α and VEGF-C protein was observed in gastric cancer.the positive expression of HIF-1α and VEGF-C in gastric cancer may play an important role in the processes of carcinogenesis,invasion and metastasis.The positive expression of HIF-1α protein was significantly correlated with the positive expression of VEGF-C protein in gastric cancer.For predicting lymph node metastasis of gastric cancer,HIF-1α and VEGF-C are of some value.
