摘要 目的:通过体外组织培养,研究小鼠眼睑体外生长发育情况。方法:剖宫产取18.5dpc胎鼠,分别切取正常彼此融合的上下眼睑及开裂的眼睑,在琼脂小岛上做组织培养,动态观察眼睑外形变化,并作HE染色观察。结果:18.5dpc胎鼠正常眼睑经过14d的培养未发生分离,睑裂沟处的组织松散,细胞凋亡呈无序性;开裂眼睑上皮细胞增殖受阻,周皮与间质部位发生分离。结论:琼脂小岛法体外培养小鼠眼睑,其组织结构及细胞增殖与迁移都不能完全重现体内情况,可能还需要添加多种生长因子。 Objective:The eyelids of 18.5dpc B6 and B6-Co mice were cultured in vitro in order to study morphological and historical dynamic change.Methods:Both the fused eyelids of B6 mice and dehiscent eyelids of B6-Co mice of 18.5dpc were obtained by caesarean operation,then cultured on agar islands and stained by using HE method.Results:Fused eyelids of B6 mice were not reopened after 14days culture and cells in the junctional zone of the eyelids of the mice in vitro showed the apoptotic changes not in order as those in vivo;epithelial cells were held-up in dehiscent eyelids of B6-Co mice,with perithelium and mesenchyme being separated simultaneously.Conclusion:The structure and cell movement could not completely reflect the fact of the eyelids developed in vivo as cultured on agar islands,and various growth factors essential should be added in the culture medium.
