子宫内膜癌组织中YKL-40、NF-κB的表达及临床意义 被引量:10

摘要 目的检测子宫内膜癌组织中人类软骨糖蛋白39(YKL-40)、核转录因子(nuclear factor kappa B,NF-κB)的表达及临床意义。方法采用免疫组织化学二步法检测40例正常子宫内膜、65例子宫内膜癌组织标本的YKL-40及NF-κB-P65的蛋白表达水平。结果 YKL-40在子宫内膜癌与正常子宫内膜中的表达率分别为76.9%和32.5%,两组比较差异有显著意义(P<0.01)。YKL-40表达与组织学分级有关,而与肌层浸润、年龄、FIGO分期、淋巴转移无相关性。NF-κB/P65在子宫内膜癌与正常子宫内膜表达阳性率为73.8%和为30%,两组比较差异有显著意义(P<0.05)。NF-κB-P65表达与组织学分级有关,而与肌层浸润、年龄、FIGO分期、淋巴转无相关性。在子宫内膜癌中YKL-40与NF-κB-P65的表达呈正相关(r=0.338,P<0.05)。结论 YKL-40、NF-κB-P65的异常表达可能在子宫内膜癌的发生、发展过程中起着重要作用,联合检测YKL-40和NF-κB-P65对子宫内膜癌的恶性程度及预后判断具有重要的临床意义。 Objective To nuclear factor- kappaB( NF- kB) detect the expression and clinical in endometrial cancer. Methods significance of YKL- 40 and Immunhistochemical method was used, the expressions of YKL - 40 and NF - kB were examined in 65 endometrial carcinoma samples, 20 normal proliferative phase endometeium and 20 normal secretary phase endometeiums (control). Results The positive expression rates of YKL- 40 in the endometrial carcinomas and the control group were 76.9 % and 32.5 % respectively, and the difference between the two groups was significant (P 〈 0.01). The expression of YKL- 40 was significantly correlated with cell differentiation, but not with clinical stages, histological type, lymphnode metastasis, ages, and myometrial invasion of endometrial carcinoma. The positive expression rates of NF - kB in the endometrial carcinomas and the control group were 73.8 % and 30 % respectively, and the difference between the two groups was significant (P〈0.05). The expression of NF- kB was significantly correlated with cell differentiation, but not with clinical stages, histological type, lyrnphnode metastasis, ages, and myometrial invasion of endometrial carcinoma. There was a positive correlation between YKL - 40 and NF -kB - P65 expressions in endometrial carcinoma (P 〈 0.05). Conclusion The Expression of YKL - 40 and NF -kB - P65 are closely related to the carcinogenesis and development of the endometrial carcinoma tumorigenesis. Combined detection of YKL- 40 and NF- kB- P65 may be helpful to evaluate malignant degree and prognosis in endometrial carcinoma.
