大肠癌患者手术前后和复发转移不同时期外周血MDR-1基因表达的研究 被引量:2

摘要 目的探讨大肠癌患者术前、术后和复发转移时外周血MDR-1mRNA的表达及其意义。方法选取2009年7月至2010年5月福建省立医院收治的术前(n=30)、术后(n=33)和复发转移(n=28)大肠癌患者,采用实时荧光定量PCR法分别检测不同时间点患者的外周血MDR-1 mRNA并进行比较。结果 MDR-1基因和内参β-actin基因扩增良好,扩增效率分别为92.8%和105.7%。实时定量PCR检测显示MDR-1基因扩增产物具有高度特异性。大肠癌术前患者外周血中MDR-1mRNA相对表达量为0.565±0.069,术后患者为0.271±0.051,差异有统计学意义(P<0.05);复发转移大肠癌患者MDR-1mRNA相对表达量为1.458±0.254,明显高于在术前及术后大肠癌患者中的表达,差异有统计学意义(P<0.05)。结论大肠癌患者外周血MDR-1 mRNA表达水平在不同时间点存在差异,不同时间点检测外周血MDR-1 mRNA表达对制定临床治疗策略有指导意义。 Objective To investigate the expression of MDR-1 mRNA in peripheral blood of colorectal cancer patients before and after operation,or with recurrence and metastasis,and analyze the differences among the expressions. Methods From July 2009 to May 2010,30 colorectal cancer patients before operation,33 patients after operation,28 patients with recurrence and metastasis were hospitalized in Fujian Provincial Hospital.Real-time fluorescent quantitative PCR(QPCR) was used to detect MDR-1 mRNA in peripheral blood of colorectal cancer patients mentioned before. Results The amplifications of MDR-1 gene and β-actin were good,and amplification effects were 92.8% and 105.7% respectively.QPCR showed that amplification of MDR-1 gene had highly specific character. The relative quantity of MDR-1 mRNA in 30 cases before operation was 0.565±0.069,and in 33 cases post-operation was 0.271±0.051,showing significant differences(P〈0.05).The relative quantity of MDR-1 mRNA in 28 cases with recurrence and metastasis was 1.458±0.254,which was higher than those in patients before and after operation(P〈0.01). Conclusion Expressions of MDR-1 mRNA in peripheral blood of patients are different in dissimilar periods of colorectal cancer.Expressions of MDR-1 mRNA will play an important role in strategy of clinical treatment.
