利用ITS和SRAP分子标记对黑龙江省野生黑木耳菌株进行遗传多样性分析 被引量:8

摘要 利用ITS和SRAP标记对黑龙江省境内采集的14株野生黑木耳菌株和一个南方栽培菌株进行遗传多样性分析。ITS序列分析结果显示:供试菌株的ITS序列变异程度较小,仅存在碱基的变化,无区域长度变异,菌株之间的遗传差异较小。利用8对引物对15个供试菌株进行SRAP扩增,共得到215条条带,其中155条为多态性条带,多态性比率为72.1%,表明黑龙江省境内野生黑木耳遗传多样性较为丰富。 To assess the genetic polymorphism of wild Auricularia auricular strains isolated from Heilongjiang Province,two DNA marker systems,the internal transcribed spacer(ITS) sequence analysis and sequencerelated amplified polymorphism(SRAP) technique were used to detect polymorphism among 15 strains.There were only few base variances in ITS region among the tested strains and the length of all strains were identical.Eight combinations of primers were selected to suvery the genetic diversity within a collection of 15 strains by sequence-related amplified polymorphism analysis which produced 215 repeatable DNA bands,of which 155 were polymorphic,accounting for 72.1%.The results showed there were more genetic information in SRAP than that of ITS,and the genetic polymorphism in Heilongjiang Province was more abundant.
