窖蛋白-1与胰腺癌关系的研究进展 被引量:1

Research progress on Caveolin-1 in pancreatic cancer

摘要 在恶性肿瘤的病死率中,胰腺癌居第4位,患者的5年生存率低于5%。由于其对手术、放疗和化疗等传统治疗方法抵抗性极强,因此治疗和预后均不理想,寻找高特异性和敏感性的治疗靶点具有重要意义。窖蛋白-1是胞膜窖中重要的结构组成部分,它和一些功能蛋白相互作用,调节细胞信号转导。本文将对窖蛋白-1与胰腺癌的研究进展作一综述,探讨窖蛋白-1在胰腺癌的增生、侵袭、转移和凋亡的过程中所发挥的作用。研究表明,窖蛋白-1在肿瘤发生发展的不同阶段,与多种信号通道相互作用,在胰腺癌细胞的增生、凋亡和转移等一系列过程中发挥了抑制或者促进的作用。 Pancreatic cancer is the fourth most common cause of cancer death, with a 5- year survival rate of less than 5%. The management and prognosis of the patients have remained dismal due to higher resistance of cancer cells to conventional approaches including surgery, radiation and chemotherapy. Therefore, there is a need for development of specific and sensitive tumor marker for pancreatic cancer. Caveolin- 1 is an essential constituent of caveolae and interacts with a variety of cellular proteins and regulates cellsignaling e- vents. In this review, we survey the functional roles of caveolin- 1 in pancreatic cancer and argue that caveo- lin-1 regulates multiple cancer-associated processes including tumor growth, cell invasion, metastasis, cell apoptosis. However, Caveolin-1 has been reported to impact both positively and negatively on various aspects of pancreatic cancer progression. The function of caveolin- 1 is interdependent on tumor stage and the expression of some signaling pathways that impact on its role during tumor progression.
