摘要
目的研究螺内酯对Wistar大鼠主动脉粥样硬化斑块及斑块内血管紧张肽转化酶2(ACE2)表达的影响。方法雄性Wistar大鼠30只随机分三组:螺内酯组、生理盐水组和空白对照组。采用高脂饮食加大剂量维生素D,腹腔注射法制备大鼠主动脉粥样硬化斑块模型。喂养50mg/(kg·d)螺内酯42d后取主动脉,苏丹Ⅳ染色确定主动脉粥样硬化斑块,病理切片观察动脉粥样硬化斑块,细胞原位杂交技术(ISH)观察斑块内ACE2mRNA表达。结果螺内酯组动脉粥样硬化斑块占主动脉内膜比率显著低于生理盐水组(螺内酯20.1%,生理盐水组33.8%,P〈0.0t)。螺内酯组主动脉粥样硬化斑块纤维帽厚度显著大于生理盐水组(螺内酯组27.3μm,生理盐水组6.3μm,P〈0.01),斑块内脂质池厚度小于生理盐水组(螺内酯组27.5μm,生理盐水组38.8μm,P〈0.05)。40倍视野下泡沫细胞数螺内酯组低于生理盐水组(螺内酯组16.5个/视野,生理盐水组43.2个/视野,P〈0.01)。螺内酯组斑块内ACE2表达高于对照组。结论螺内酯能延缓大鼠主动脉粥样硬化斑块进展并增加斑块稳定性及主动脉粥样硬化斑块内ACE2表达。
Objective To study the effect of spirolactone on mice aortic atherosclerosis plaque and ACE2 express in the plaque. Methods Thirty male wistar mice were randomly divided into three groups: spirolactone group, saline group and control group. Establishing rat aortic atherosclerosis model using large amount vitamin D3 intraperitoneal injection and high cholesterol diet. Decollement aorta after feed the mice with spirolactone 50 mg/( kg·d) for 42 days, the aorta was stained with Sudan IV to observe the atherosclerosis plaque. Pathological slice to observe the atherosclerosis plaque. In situ hybridization( ISH ) to observe ACE2 mRNA express in the plaque. Results The percentage of atherosclerosis plaque in the aorta intima area of spirolactone group was obviously less than the saline group(20.1% vs 33.8, P 〈 0. 01 ). In spirolactone group , the fibrous cap of the atherosclerosis plaque was thicker than the saline group(27.3μm vs 6.3 μm, P 〈 0. 01 ) while the lipid pool was thinner than the saline group(27.5 vs 38.8, P 〈0.05). The spirolactone group had more foam cells than the saline group in the 40 x microscope view( 16.5/FN vs 43.2/FN, P 〈0.01 ). The ACE2 express in the plaque of spirolactone group was more than saline group. Conclusion Spirolactone can obviously improve the mice aorta atherosclerosis plaque and increase the stabilization of the plaque, and it can also increase the ACE2 expression in the aortic atherosclerosis plaque.
