摘要
以猪、牛、羊、鸡等动物新鲜冷冻样品为实验材料,采用SDS法和异硫氰酸胍法提取动物肌肉组织基因组DNA,对提取的DNA进行光密度、琼脂糖凝胶电泳和PCR检测,比较了这两种方法的提取效果。结果表明:采用SDS法和异硫氰酸胍法均能从动物肌肉组织提取到完整的基因组DNA,均可满足PCR等后继分子生物学实验的要求。但是异硫氰酸胍法提取的基因组在纯度和浓度及稳定性方面优于SDS法,且操作简单,耗时短,更利于快速检测。
Genomic DNA of musclar tissues from swine,cattle,goat and chicken were extracted through SDS method and GuSCN method,respectively. The DNA was detected by optical density,agarosegel electrophoresis and polymerase chain reaction. The extraction effects of two methods were compared. Results indicated that high quality genomic DNA was separately extracted from muscle tissue of 4 animals by the SDS method and the GuSCN method,and meeted the requirements of varieties of molecular biology experiments. But the GuSCN method improved yield and purity of DNA,and had several advantages,such as simply manipulate,consume time shortly,and so on,which was most fit for extraction total DNA of animal muscle tissue.
