竹红菌素脂质体的制备及其包封率测定方法的研究 被引量:1

摘要 采用薄膜分散-均质法制备了竹红菌素脂质体,采用TEM观察脂质体形貌,用Zeta电位及粒度分析仪检测脂质体粒径,并选择石油醚萃取法、大孔树脂柱层析法和微柱离心法分别测定竹红菌素脂质体包封率。结果表明,该脂质体呈椭球形,其粒径分布范围为118.2~578.0 nm,平均粒径为261.4 nm;大孔树脂柱层析法最适合该体系包封率的测定,脂质体包封率为42.84%,竹红菌素总回收率为91.28%。 Hypocrellin liposome was prepared by thin - film dispersion - homogenization method. TEM was used to observe its morphology, and its particle size was measured with zeta potential and particle size analyzer. Petroleum ether extraction method, macroporous resin column chromatography and micro - column centrifuge method were respectively adopted to separate unencapsulated hypocrellin and liposome thus determine its extent of encapsulation. Result showed that, hypocrellin liposome is ellipsoid - shaped with particle size ranged in 118.2 -578.0 nm and the mean particle diameter is 261.4 nm;macroporous resin column chromatography is the most suitable method to determine the extent of encapsulation of the system, which reveals that the extent of encapsulation achieves 42.84% and 'the total recovery of hypocrellin achieves 91.28%.
