摘要
目的探讨多肽P110联合顺铂对不同肿瘤细胞的杀伤效应及其对顺铂的可能增敏作用。方法采用胃癌细胞SGC-7901、结肠癌细胞HCT-116、宫颈癌细胞HeLa、肺癌细胞A-549、人脐静脉血管内皮细胞(HUVEC)细胞系。MTT法检测顺铂(DDP)组、P110组及顺铂+P110组对上述细胞生长的影响,及P110对顺铂在不同肿瘤细胞中的增敏作用。结果 P110或顺铂对不同肿瘤细胞的增殖均有明显抑制作用,且呈浓度依赖性;多肽P110联合不同浓度顺铂对不同肿瘤细胞的抑制率均高于顺铂组,差异有统计学意义(P<0.05),其中,P110联合10μmol.L-1顺铂杀伤HCT-116和HeLa细胞的效果与高浓度顺铂(30、90μmol.L-1)相当,且对HUVEC细胞的毒性作用较小;在相同浓度梯度下,P110联合顺铂对宫颈癌HeLa细胞的杀伤作用均优于其他肿瘤细胞(P<0.01)。结论多肽P110可以增强顺铂抗肿瘤作用,联合用药可以减少顺铂的用量,减轻对HUVEC的毒性作用;多肽P110单独使用亦有一定的杀伤作用;联合用药时对宫颈癌HeLa细胞的杀伤效果最好。
Aim To study the cytotoxic effect of polypeptide P110 combined with cisplatin on cancer cells and the probable sensitizing effect on cisplatin.Methods Cell lines of SGC-119,HCT-119,HeLa,A-549,HUVEC were used.MTT was used to measure inhibitory effects on the growth of SGC-119,HCT-119,Hela,A-549,HUVEC cells.Results Cisplatin(or P110) dose-dependently inhibited proliferation of SGC-7901,HCT-116,HeLa and A-549.The inhibitory rate of P110 combined with cisplatin(3.3,10,30,90 μmol·L-1)was more than that of cisplatin single group(P0.05);10 μmol·L-1 cisplatin combined with P110 could achieve the greatest anti-tumor effect on HCT-116 and HeLa cells,and least toxicity on HUVEC cells.At the same concentration gradient,the cytotoxic effect on Hela was better than others(P0.01).Conclusions Polypeptide P110 can enhance the cytotoxic effect of cisplatin on gastric cancer cell line SGC-119,colon cancer cell line HCT-119,cervical carcinoma cell line HeLa and lung cancer cell line A-549.Polypeptide P110 alone has the cytotoxic effect on cancer cells too.Cisplatin combined with P110 can reduce the dose of cisplatin,and cut down the toxicity on HUVEC cells.The cytotoxic effect on cervical carcinoma cell line HeLa is the best.
