摘要
目的研究菘蓝种子总多酚的最佳提取工艺条件,并初步探讨其抗氧化活性。方法采用单因素试验和L9(34)正交试验法,确定菘蓝种子总多酚最佳提取工艺;并考察其对DPPH.的清除能力。结果菘蓝种子总多酚的最佳提取工艺为:料液比1︰10,甲醇质量分数60%,提取温度50℃,提取时间40 min。抗氧化活性评价显示菘蓝种子总多酚清除DPPH.的能力呈浓度依赖效应,当总多酚质量浓度为1.0μg/mL时,与BHT的抗氧化能力基本相当。结论本研究建立了菘蓝种子总多酚的最佳超声提取工艺条件,总多酚得率为(8.92±0.03)mg/g,DPPH.法评价结果显示其具有一定的抗氧化活性。
AIM To establish the optimal extraction technology for total polyphones from seeds of Isatis indigotica Fort.,and evaluate its antioxidant activities.METHODS Using the contents of total polyphones as evaluation index,the effects of solid-liquid ratio,methanol concentraction,extraction temperature and extraction time on the contents of total polyphones were discussed,respectively.Based on the results of one-factor experiment,an orthogonal test of L9(34) was corried out to obtain the optimum conditions,and the DPPH free radical scavenge activity was also discussed in this experiment.RESULTS The optimum conditions for total polyphones were as follows: solid-liquid ratio of 1∶ 10,60% methanol concentration,50°C extraction temperature;40 min extraction time,and the total polyphones showed obvious antioxidant activity.CONCLUSION Under the optimum extraction conditions,the contents of total polyphones are(8.92±0.03) mg/g.And with the increase of total polyphones concentration,their DPPH radical scavenging activity increases.
