摘要
目的观察仙鹤草降糖作用,并从氧化性损伤角度初步探讨其作用机制。方法雄性SD大鼠腹腔注射链脲佐菌素建立糖尿病大鼠模型。将造模成功大鼠随机分为糖尿病模型组,仙鹤草低、高剂量组(60、120 g/kg,每天经口灌胃)及对照组。观察大鼠体重、血糖及血清胰岛素水平变化,检测血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量;做胰腺病理组织学检查。结果模型组大鼠血清胰岛素和血糖水平分别为(7.53±2.15)μIU/mL、(17.55±6.08)mmol/L,血清GSH-Px活性和MDA含量分别为(1110.75±268.16)μmol/mL、(6.10±0.88)nmol/mL;低、高剂量仙鹤草组胰岛素和GSH-Px活性分别为(16.79±6.81)、(18.29±7.15)μIU/mL和(1264.50±273.20)、(1824.38±240.59)μmol/mL,明显高于模型组(P<0.01),血糖水平和MDA含量分别为(4.71±2.19)、(3.92±1.17)mmol/L和(4.99±0.90)、(4.92±0.50)nmol/mL,明显低于模型组(P<0.01)。结论仙鹤草可通过增加GSH-Px活性和降低MDA含量发挥抗糖尿病作用。
Objective To study the hypoglycemic effects of agrimony and to explore its mechanism.Methods The streptozotocin(STZ)-induced diabetic rats were randomly divided into diabetic model group,low dose agrimony group(60 g/kg),high dose agrimony group(120 g/kg),and a control group was set additionally.After 4 weeks of treatment,the effects of agrimony on weight,blood glucose and insulin levels of diabetic rats were observed and the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and the contents of maloraldehyde(MDA) in plasma were measured with chromatometry,and the pancreas were excised and observed under microscope.Results The serum insulin and glucose levels of diabetic rats were 7.53±2.15 μIU/ml and 17.55±6.08 mmol/L;the activities of GSH-Px and the contents of MDA of diabetic rats were 1 110.75±268.16 μmol/ml and 6.10±0.88 nmol/ml.Administration of agrimony decreased fasting blood glucose and the content of MDA in plasma efficiently and increased insulin level and the activity of GSH-Px in diabetic rats.The serum insulin of agrimony groups(60g/kg,120g/kg) were 16.79±6.81 μIU/ml and 18.29±7.15 μIU/ml(P0.01);the glucose were 4.71±2.19 mmol/L and 3.92±1.17 mmol/L(P0.01);the activities of GSH-Px were 1264.50±273.20 μmol/ml(P0.05) and 1824.38±240.59 μmol/ml(P0.01);the contents of MDA were 4.99±0.90 nmol/ml(P0.05) and 4.92±0.50 nmol/ml(P0.01).Conclusion Agrimony has anti-diabetes effect,which is probably due to the increase of GSH-Px activity and the decrease of MDA content.
