摘要
目的建立同时测定桂附地黄丸中没食子酸、5-羟甲基糠醛、马钱苷、芍药苷和丹皮酚5种成分的反相高效液相色谱分析方法。方法色谱柱为Scienhome Kromasil C18柱(250 mm×4.6 mm,5μm),流动相A为体积分数为0.1%的磷酸溶液、B为体积分数为0.1%的磷酸甲醇溶液,梯度洗脱,柱温(30.00±0.15)℃,检测波长为190~400 nm DAD检测。结果没食子酸、5-羟甲基糠醛、马钱苷、芍药苷和丹皮酚分别在0.06~1.60、0.06~1.60、0.05~0.60、0.05~0.60和0.12~3.20 g.L-1内峰面积与质量浓度呈良好的线性关系。5种成分的平均回收率分别为98.7%、101.2%、99.0%、98.5%和99.6%。结论该方法可用于桂附地黄丸的质量控制。
Objective To establish an RP-HPLC method for the simultaneous determination of the five components in Guifudihuang pills (GFDHP, traditional Chinese medicines). Methods GFDHP was separated by RP-HPLC on Scienhome Kromasil C18 (250 mm ×4.6mm,5μm)column with the gradient elution solvent composed of 0. 1% (φ) phosphoric acid water and 0. 1% ( φ ) phosphoric acid methanol. The flow rate was 1.0 mL-min-1 ,the column temperature was maintained at(30.00 ±0. 15 )℃ ,and ard UV detection wavelengths were set from 190 to 400 nm to determine the contents of the five components. Results Good linear relationships were obtained between peak area and concentrations of the five components, and the calibration curves were linear in the ranges of 0. 06-1.60,0.06-1.60,0. 05-0. 60,0. 05-0.60,0. 12-3.20 g.L-1 for the gallic acid,5-hydroxymethyl-2-furaldehyde, loganin, paeoniflorin and paeonol, respectively. The average recoveries were 98.7%, 101.2% ,99.0% ,98.5% and 99.6% ,respectively. Conclusions This method is simple, and accurate. It can be used for the quality control of the GFDHP.
