摘要
以油脂加工企业热榨油后产生的花生粕为原料,研究花生多糖纤维素酶法提取工艺。在单因素试验的基础上,选定提取时间、提取温度、pH值和酶添加量等4因素实施中心组合试验设计,建立多糖提取率的二次回归方程,通过响应面分析得到优化组合条件。结果表明,在提取时间253min、提取温度43℃、pH值为5.9、酶添加量为0.3%的条件下,花生多糖的提取率达到极值为6.77%。为了检验模型预测的准确性,在优化的条件下进行提取试验,花生多糖提取率为6.47%。
Crude polysaccharides were prepared by enzymatic method from the residue of peanut after lipid extraction. According to single-factor ex- periments, four factors including extraction time, extraction temperature, pH values and cellulase concentration were selected for the central compos- ite design to establish a quadric regression equation for predicting the yield of polysaccharides. By response surface analysis, the optimum extraction conditions were obtained as follows: extraction time 252min, extraction temperature 43℃, pH value 5.9 and the amount of cellulase 0.3%, which led to prediction and measured values ofpolysaccharides yield to 6.77% and 6.47%, respectively.
