摘要
目的探讨原花青素对SD大鼠大脑中动脉缺血再灌注损伤的保护作用。方法采用线栓法建立大鼠大脑中动脉缺血再灌注模型,将SD大鼠60只随机分为正常对照组、模型组、阳性对照组、原花青素高、中、低剂量组(100,200,400 mg/kg),观察原花青素对大鼠神经行为学评分,脑梗死体积,脑组织匀浆丙二醛(MDA)、超氧化物歧化酶(SOD)水平变化,及caspase-3表达的影响。结果原花青素能明显减少大鼠脑栓塞模型的梗死体积,降低神经行为学评分和MDA含量,提高SOD活性,下调caspase-3的表达。结论原花青素对脑缺血再灌注损伤具有一定的保护作用,其作用机制可能与其抗氧化活性以及下调caspase-3的表达有关。
Purpose To study the neuroprotective effects of pro cyanidins on rat brain after cerebral ischemia-reperfusion. Methods Sixty healthy SD rats were randomly assigned into the sham operative group, focal cerebral isehemia group, nimodipine group and three treatment groups, which with 100, 200 and 400 mg/kg procyanidins respectively. After gastric gavage of PC for 7 days, SD rat models of cere- bral embolism were established. The neurological function was evaluated and the activity of superroxide dismutase ( SOD ) and the content of malondialdehyde (MDA) in brain tissue was measured after 24 h focal cerebral ischema cerebral pathological changes were observed by TTC staining. By using immuno- histochemieal SP methods, we detected the number of easpase-3 positive cell. Results Proanthocyanidin can significantly reduce the neurological deficits scores, inhibited cerebral infarction volume in rats with cerebral embolism, decreased MDA level and increased SOD activity, down-regulated caspase-3 expres- sion. Conclusion Proantboeyanidin has protective effect against cerebral isehemia-reperfusion, and its mechanism may be associated with anti-oxidization and down-regulated caspase-3 expression.
