摘要
从腐烂大枣中筛选到1株产纤维素酶的菌株ZJ01,利用ITS序列进行分子鉴定,该菌株属于围小丛壳(Glomerella cingulata),为大枣炭疽病菌。对菌株ZJ01所产羧甲基纤维素酶(CMCase)、滤纸酶(FPase)和β-葡萄糖苷酶(BG)等3种纤维素酶的最适pH值、最适反应温度进行了分析,并比较了不同pH值、温度、金属离子等对3种纤维素酶活性的影响。采用CMC-SDS-PAGE技术对CMC酶进行活性染色,发现3个透明条带,分子量大小在31.0~42.7 ku之间,结果表明该菌种能分泌3种类型的内切纤维素酶。
A fungus strain ZJ01 producing cellulases was isolated from rotted Chinese date.This strain was identified as Glomerella cingulata by molecular biological method according to ITS(internal transcribed spacer) sequence.The optimum pH value and temperature of cellulases including endoglucanase(CMCase),exoglucanase(FPase) and β-glucosidase(BG) were studied.Effects of different pH,temperature and metal ions on the activities of the three cellulases were also analyzed.Activity staining of CMCase were performed after CMC-SDS-PAGE.Three bands were observed and their molecular weights were between 31.0 ku and 42.7 ku.These results indi-cated that the ZJ01 strain could secret three different types of endoglucanases.
