摘要
[目的]分析复发鼻咽癌和初发鼻咽癌的蛋白质磷酸化表达差异,以期为复发鼻咽癌的发生机制提供分子依据。[方法]选择4例复发鼻咽癌患者和4例初发鼻咽癌患者的组织标本,实验室提取总蛋白,应用磷酸化抗体蛋白质芯片与提取蛋白进行杂交,检测两者之间的差异磷酸化位点。通过phosphosite在线网络数据库分析蛋白质磷酸化位点变化及在信号通路中的作用。[结果]复发鼻咽癌和初发鼻咽癌的蛋白质磷酸化表达不同,共筛选出16个差异磷酸化位点,15个差异蛋白,复发鼻咽癌组织中上调蛋白8个,下调蛋白7个。[结论]初发鼻咽癌和复发鼻咽癌组织之间存在蛋白质磷酸化水平差异,可能与鼻咽癌复发机制有关。
[Purpose] To investigate the differential expression of phosphorylation of protein chip in relapsed nasopharyngeal carcinoma (rNPC) and primary nasopharyngeal carcinoma(pNPC), to provide molecular evidence of occurrence mechanism for recurrent NPC. [Methods] Four cases with pNPC and 4 cases with rNPC were enrolled. Total protein was analyzed by antibody microarray. The differential phosphorylation sites of rNPC and pNPC were detected by phospho-antibody protein chip hybridized with the extracted proteins. The change and role of phosphorylation site in the signaling pathway was analyzed by phosphosite online network database. [Results] rNPC and pNPC had differential phosphorylation level of proteins,and with 16 differentially phosphorylation site and 15 expressed proteins were identified. Eight proteins up regulation and 7 proteins down regulation were found in rNPC.[Conclusion] Protein phosphorylation level is different between pNPC and rNPC, it might relate to the occurrence of rNPC.
