摘要
目的: 为阻止孕期饮酒所致胚胎生长发育迟缓和发育异常的作用, 本研究采用全胚胎培养法研究具有多种防御功能的Vc对酒精发育毒性的拮抗作用。方法:孕9.5 天大鼠胚胎施与不同剂量的酒精和Vc, 观察Vc对酒精的拮抗作用,并应用2×3 析因分析判断联合作用类型。结果: 酒精浓度为2000m g/L时, Vc能使胚胎发育有所改善, 但与酒精单独作用时无显著性差异(P> 0.05)。酒精浓度为4000m g/L时, 10μg/m LVc可拮抗酒精的发育毒性作用, 胚胎体长、DNA含量、蛋白质含量与卵黄囊直径均与同剂量的酒精单独作用时有显著性差异 (P< 0.05)。翻转、体长、DNA 含量有交互作用。对有交互效应的指标进一步分析发现, 酒精浓度为4000m g/L时, Vc对酒精发育毒性呈拮抗作用。
Objective:In order to prevent the effects of developmental retardation and abnormalities induced by drinking alcohol during pregnancy,this study used whole embryo culture(WEC) to research the antagonism of developmental toxicity of alcohol by vitamin C(Vc),which possesses multiple function.Method:The embryos of 9 5days were administrated the different doses of alcohol and Vc,and at the same time,factorial experimental analysis of 2×3 was used to analyze the types of combined action.Result:The result showed that when alcohol concentration was 2000mg/L,Vc(10μg/ml) could partly improve development of embryo,but it had no significant difference.Vc(10μg/ml) could antagonize developmental toxicity of alcohol of 4000mg/L,and crown rump length,the confents of DNA and protein,and diameter of visceral yolk sac(VYS) had significant differences compared with alcohol independent action.By means of factorial experimental analysis,it was shown that there were some interfactions between crownrump length,the content of DNA,and turn location.Analysis of interaction showed that Vc could antagonize developmental toxicity induced by alcohol.This result hints that Vc has only a partial action to embryo growth improvement.
