摘要
【目的】研究发情周期各阶段KiSS-1/GPR54系统在母羊下丘脑中的表达规律。【方法】将12只雌性小尾寒羊随机分为4组,每组3只,分别于发情前期、发情期、发情后期和间情期,颈动脉放血致死后采集其下丘脑,采用实时荧光定量PCR技术研究母羊发情周期不同阶段下丘脑中KiSS-1和GPR54基因的表达规律。【结果】母羊发情周期各阶段下丘脑上均有KiSS-1和GPR54基因表达;发情期母羊下丘脑KiSS-1基因的表达量极显著高于发情后期、间情期和发情前期(P<0.01),而发情后期、间情期和发情前期之间差异不显著;发情周期的不同阶段,GPR54基因在母羊下丘脑中的表达量无显著差异(P>0.05)。【结论】发情期母羊下丘脑KiSS-1基因的表达量极显著高于发情周期其他阶段(P<0.01),GPR54基因在母羊发情周期各阶段的表达量无显著差异。
【Objective】 The study investigated the expression of KiSS-1/GPR54 system in hypothalamus of ewes during the estrous cycle.【Method】 Twelve female Small Tail Han sheep were divided into four groups of 3,as the proestrous,estrous,metestrous and diestrous.All the ewes were killed by carotid artery bloodletting and then hypothalamus were collected for the evaluation of expression of KiSS-1 and GPR54 gene in hypothalamus by time fluorescence quantitative PCR method.【Result】 Results showed that both KiSS-1 and GPR54 gene expressed in ewe hypothalamus during the estrous cycle and expression of KiSS-1 gene in ewe hypothalamus changed with the estrous cycle,i.e.,with higher level in the estrous than other stages(P0.01).However,the expression of GPR54 gene in hypothalamus did not show significant difference during the estrous cycle(P0.05).【Conclusion】 Expression of KiSS-1 gene in hypothalamus of ewe during the estrous was higher than that of other stages of the estrous cycle(P0.01),however,the expression of GPR54 gene in hypothalamus did not show significant difference during the estrous cycle(P0.05).
