PPARγ配体曲格列酮对绒癌JEG-3细胞浸润能力的影响

摘要 目的探讨过氧化物酶体增殖物激活型受体γ(PPARγ)及配体曲格列酮(Troglitazone,TGZ)对绒癌JEG-3细胞浸润能力的影响。方法通过免疫荧光细胞化学方法检测JEG-3细胞系中PPARγ的表达,采用Transwell侵入系统检测PPARγ的激动配体TGZ对绒癌细胞浸润能力的影响。结果在绒癌JEG-3细胞系中有PPARγ蛋白表达,PPARγ激动配体TGZ处理后浸润人工基底膜细胞数明显减少,与对照相比差异有统计学意义(P<0.01),且具有剂量依赖关系。结论 PPARγ被配体TGZ激活后抑制绒癌JEG-3细胞浸润能力,这为PPARγ配体在绒癌治疗中可能的临床应用提供理论和实验基础。 Objective To investigate the effect of PPARγ ligand Troglitazone(TGZ) on invasion of JEG-3 cell from chorioepithelioma.Methods We examined the expression PPARγ in JEG-3 cell by immunocytochemistry.We next examined,using the cytotrophoblast culture model,the effect of PPARγ ligands Troglitazone on invasion of JEG-3 cell in vitro.Results There was the expression of PPARγ protein in JEG-3 cell,and mainly localized in the nucleus of cell.After treatment with TGZ,cells of invision artificial basement membrane were decreased obviously in a concentration dependent manner,and difference was significant compared with control(P0.01).Conclusion PPARγ ligand TGZ inhibit the invision of chorioepithelioma JEG-3 cell,and these provide the theoretical and experiment basement for potential clinical application of PPARγ ligands in the therapy of chorioepithelioma.
