3-硝基-1,2,4-三唑-5-酮对大鼠亚急性毒性实验研究 被引量:2

Experimental study on the sub-acute toxicity of 3-nitro-1,2,4-triazol-5-one in rats

摘要 目的探讨新型含能材料3-硝基-1,2,4-三唑-5-酮(3-nitro-1,2,4-triagol-5-one,NTO)的亚急性经口毒性效应,为亚慢性毒性或慢性毒性实验提供剂量设定依据。方法按照《化学品毒性鉴定技术规范》,选择健康成年SD大鼠44只,雌雄各半,随机分为对照组、NTO 250、500和1 000 mg/kg染毒组。染毒组用NTO-植物油混悬液灌胃(10 ml/kg),对照组用等量植物油灌胃,经口连续灌胃28 d。观察大鼠体质量、脏/体比;分析红细胞、白细胞计数等10项血常规指标;血清天冬氨酸转氨酶(AST)、尿素氮、肌酐(Cr)水平等12项生化指标。取心、肝、脾等7种脏器进行病理组织学检查。结果中、高剂量组雄性大鼠体质量增长缓慢,睾丸/体比值低于对照组。高剂量组雄性大鼠平均红细胞体积升高,雌性大鼠淋巴细胞百分比降低,低、中、高剂量组血清AST活力明显升高,高剂量组血清Cr升高,与对照组比较差异均有统计学意义(P<0.05)。与对照组比较,中、高剂量组大鼠肝脏、肾脏和睾丸病理组织学检查显示发生病变 NTO对大鼠肝脏、肾脏和睾丸有一定的损伤作用。 Objective To discuss the sub-acute oral toxicity of new energetic materials 3-nitro-1, 2, 4- triazol-5-one ( NTO), for providing the setting dosage of sub-chronic toxicity or chronic toxicity. Methods According to The Technical Regu- lation of Chemicals Toxicity Identification, 44 healthy adult SD rats were randomly divided into control group and NTO exposure groups(250, 500 and 1 000 mg/kg). The exposure groups were given 10 mL/kg NTO-vegetable oil by garage for 28 days. The body weight and organ-body ratios of rats were observed, and the blood routine, serum biochemical indexes were analysed respec- tively. Seven kinds of organs for pathological histology were examined. Results In the median and high dose groups, the body weight slow growth in male rats, and testis-body ratio were lower than those of the control group. In the high dose group, the mean corpuscular volume (MCV) was increased in male rats, the percentage of lymphocytes was reduced in female rats. Com- pared with the control group,the activity of aspartate aminotransferase(AST) was increased significantly in the low, median and high dose groups ( P 〈 0. 05 ). In the high dose group, the creatinine of serum was higher than that of the control group ( P 〈 0. 05). Compared with the control group, the liver, kidney, testical of rats showed pathological changes in the median and high dose groups. Conclusion NTO could injure the liver,kidney,testical of rats.
