阿米卡星脂质体的包封率测定

Determination of the encapsulation efficiency of liposomal amikacin

目的：为了对阿米卡星（ａｍｉｋａｃｉｎ）脂质体进行质量评价，测定了阿米卡星脂质体的包封率。方法：采用新型的两步洗脱法，即前２５ｍｌ 以蒸馏水，后５０ｍｌ 改为ＮａＣｌ 溶液作为流动相进行分离。结果：本方法既可保证脂质体在凝胶柱上不被破坏，又可保证脂质体和游离药物在柱上无吸附现象，柱回收率良好。结论：本方法测定阿米卡星脂质体的包封率新颖。

OBJECTIVE:To determine the encapsulation efficiency of liposomal amikacin in order to evaluate the quality of liposomes. METHODS:A novel gel filtration chromatography with two mobile phaseelution, i.e.,the first 25ml distilled water and the following 50ml 0.9 per cent NaCl solution (saline),was used. RESULTS:This method not only protected liposomes from destroying, but also no drug adhesive in gel column. The encapsulation efficiency of liposomal amikacin was 85.3%.CONCLUSION: This analytical method to determine the encapsulation efficiency of liposomal amikacin was novel and reliable.
