摘要
以文冠果带芽茎段为外植体,以MS配方为基本培养基,采用不同激素组合对其进行离体培养试验,结果表明:6-BA1.00 mg.L-1+NAA0.20 mg.L-1和6-BA1.00 mg.L-1+NAA0.50 mg.L-1的激素组合适宜诱导芽和增殖培养,分化率分别为90.4%和89.3%;苗高分别为1.96 cm和1.97 cm;增值倍率为4.5和4.8,丛生芽粗壮。而1/2MS+IBA0.50 mg.L-1为其最适生根培养基,生根率93.1%,平均生根4.8条,平均根长1.5 cm。生根苗移栽应严格控制温湿度,从成活率和成本综合考虑,园土炉灰=21是较为适宜的移栽基质,移栽30 d,平均苗高可达4.5 cm,成活率90%以上。
Experiments of in vitro culture were conducted under different hormone combination conditions with MS as basic medium and stems with buds of Xanthoceras sorbifolia as explants.The hormone combinations of MS+6-BA1.00 mg·L-1+NAA0.20 mg·L-1 and MS+6-BA1.00 mg·L-1+NAA 0.50 mg·L-1 of were more suitable for induction(90.4% and 89.3%,respectively) and proliferation(4.5 and 4.8,respectively) of X.sorbifolia and the seedling heights were 1.96 cm and 1.97 cm,respectively.The 1/2MS+IBA0.50 mg·L-1 was the optimal rooting medium,with 93.1% rooting rate,4.8 roots and 1.5 cm root length on average.The temperature and humidity should be restrictedly controlled when seedlings were transplanted.Considering the cost and survival rate,the optimal medium for transplantation was garden soilfurnace ash at 21.After transplanted 30 d,the average height of seedlings was 4.5 cm with above 90% survival rate.
