老年低额溞卵黄蛋白的分离纯化及性质

摘要 为了研究老年低额溞卵黄蛋白(vitellion,Vn)在环境检测中的应用,采用制备电泳方法,从老年低额溞整体匀浆液提取的粗蛋白中纯化其Vn.聚丙烯酰胺凝胶电泳(PAGE)检测表明,SephadexG-100得不到高纯度的Vn,制备电泳可以得到高纯度的Vn,达到了实验要求.对老年低额溞Vn的性质进行研究,老年低额溞Vn总分子质量为389 900u,含有5种亚基,分子质量分别为76 500,63 000,59 200,52 700,47 200u,含有2个二硫键,等电点为5.0~5.5,是一种酸性蛋白. In order to study the application of Simocephalus vetulus'vitellion in the environment testing,from homogenate liquid of S.vetulus,preparative electrophoresis was used to purify vitellion.Protein was detected by polyacrylamide gel electrophoresis,it indicated that chromatography(SephadexG-100) couldn't guarantee high-purity of vitellion.Preparative electrophoresis showed that the high purity of the protein could be obtained.Properties of S.vetulus'vitellion was analysed.Molecular mass of the protein was 389 900 u.Five subunits were found and their molecular mass were 76 500,63 000,59 200,52 700,47 200 u.The protein had two disulfides.Its pI was 5.0~5.5,and it was a acidic protein.
