双黄升白颗粒对化疗所致骨髓抑制荷瘤小鼠细胞增殖的调控 被引量:1

摘要 目的:探讨双黄升白颗粒对化疗所致骨髓抑制荷瘤小鼠骨髓及肿瘤细胞增殖的调控作用。方法:本研究采用Lewis肺癌荷瘤小鼠,30只小鼠随机分为空白组、模型组和治疗组。除空白组外,腹腔注射环磷酰胺制作骨髓抑制模型。治疗组用40g/(kg.d)双黄升白颗粒治疗6 d后,常规检测血常规、有核细胞数、肿瘤大小、骨髓及肿瘤细胞周期,以RT-qPCR(reverse tran-scription quantitative real-time PCR,RT-qPCR)检测增殖相关因子C-myc、N-myc、L-myc,并以免疫组化法进行验证。结果:模型组骨髓组织的G0/G1期细胞比例均低于空白组(P<0.05),PI和C-myc、N-myc、L-myc的表达均高于空白组(P<0.05)。肿瘤组织G0/G1期细胞比例均低于空白组(P<0.05),PI和C-myc、N-myc、L-myc表达均高于空白组(P<0.05)。治疗组骨髓G0/G1期细胞比例低于模型组及空白组(P<0.05),PI及C-myc、N-myc、L-myc的表达均高于模型组及空白组(P<0.05)。肿瘤组织G0/G1期细胞比例高于空白组及模型组(P<0.05),PI及C-myc、N-myc、L-myc的表达均低于模型组及空白组(P<0.05)。结论:双黄升白颗粒对骨髓抑制Lewis肺癌荷瘤鼠的骨髓及肿瘤细胞增殖具有双向调控作用,其机制可能与调控增殖相关因子C-myc、N-myc、L-myc的表达有关。 Objective: To explore the regulation of cell cycles by Shuanghuang Shengbai Granule in Lewis-bearing mice with chemotherapy-induced mye1osuppression.Methods: Thirty Lewis-bearing mice were randomly divided into control group,untreated group and treated group.In addition to the control group,the intraperitoneal injection of cyclophosphamide produced the model of mye1osuppression.Mice in the treated group were treated with Shuanghuang Shengbai Granule for 6 days.Routine testing blood,bone marrow cells,tumor size,cell cycle progressions of cells collected from bone marrow and tumor tissues were assayed.The expressions of the proliferation-related factors such as C-myc、N-myc and L-myc were detected by reverse transcription quantitative real-time PCR(RT-qPCR)and mmunohisiochemical method.Results: Compare with the control group,the percentage of bone marrow cells in G0/G1 phase in the untreated group was decreased,the PI of bone marrow cells and the expression of C-myc、N-myc、L-my was increased(P0.05).The percentage of tumor cells in G0/G1 phase was increased,the PI of tumor cells and the expression of C-myc、N-myc、L-my was increased(P0.05).Compare with the control group and the untreated group,the percentage of bone marrow cells in G0/G1 phase in the treated group was decreased,the PI of bone marrow cells and the expression of C-myc、N-myc、L-my was increased(P0.05).The percentage of tumor marrow cells in G0/G1 phase in the treated group was increased,the PI of tumor cells and the expression of C-myc、N-myc、L-my was decreased(P0.05).Conclusion: Shuanghuang Shengbai Granule resulted in a dual control of the cell cycles in Lewis-bearing mice with chemotherapy-induced mye1osuppression by regulating the expressions of the proliferation-related factors such as C-myc、N-myc and L-myc.
