摘要
目的 :研究脑缺血海马ATP含量变化与ATP酶活性变化的关系及低温的影响。方法 :沙鼠前脑缺血再灌注模型 ,脑缺血时间 10分钟。测定脑缺血 10分钟、再灌注 3 0分钟和 60分钟时海马突触体ATP和ATP酶活性的变化。结果 :脑缺血后ATP和ATP酶活性明显下降 (P <0 .0 1)。再灌注后ATP含量有明显恢复 ,但ATP酶活性恢复的程度明显小于ATP恢复的程度 (P <0 .0 5 )。低温组ATP和ATP酶活性恢复的程度均高于常温组 (P <0 .0 5 )。结论 :脑缺血后应用低温治疗可促进海马突触体ATP和ATP酶活性的恢复 ,这可能为其减轻脑缺血再灌注损伤的机制之一。
Objective:TostudytherelationshipbetweenthechangesofATPcontentandATPaseactivitiesaftercerebral ischemia.Methods:Theischemiawasinducedbyocclusionbothcommoncarotidarteriesfor 10minutesingerbils.ATP contentandNa+,K+ ATPaseactivitywereassasyedfollowingischemiaandreperfusioningerbils.Results:TheATPcon tentofthehippocampusinnormothermicreperfusion (3 7℃ )groupafterischemiawasremarkablylowerthanthatinsham operatedgroup .CerebralischemialedtoasignificantreductionofNa+,K+ ATPaseactivityinnormothermicreperfusion group .BothATPcontentandATPaseactivityinhypothermic (3 2℃ )groupwerehigherthanthoseinnormothermicgroupfollowing 60minutesreperfusion .ThechangesofCa2 + ATPaseactivityweresimilartothatofNa+,K+ ATPase.Conclu sion :ItissuggestedthathypothermictreatmentcouldleadtoquickerrecoveryofATPcontentandATPaseactivityduringis chemiaandreperfusion ,whichcouldbeoneofthemechanismsreducingischemia/reperfusioninjury .
