脑胶质瘤中性鞘糖脂抗原的研究 被引量:3

摘要 目的 为了解脑胶质瘤中性鞘糖脂(NGSLs) 表达的规律及发现脑胶质瘤相关胚胎鞘糖脂抗原。 方法 本实验运用改良的Hakomori 的方法提取、纯化了脑胶质瘤及胚胎脑的中性鞘糖脂,并行高效薄层层析(HPTLC) 。 结果 胶质瘤的NGSLs 以CDH( 二己糖神经酰胺)及CTH( 三己糖神经酰胺) 的表达较正常人脑强,胚胎脑NGSLs 以CMH(单己糖神经酰胺)及CTH表达为主,与正常人脑组织NGSLs 相比,胶质瘤及胚胎脑均不含有Globoside( 红细胞糖苷脂) 。 结论 脑胶质瘤及胚胎脑的NGSLs 表达具有一定程度上的相似性。 PURPOSE The expression of the neutral glycosphingolipids in gliomas and the glioma associated embryonic N GSLs antigen was studied.METHODS A modified Hakomori′s method was used to extract and purify the N GSLs from glioma and embryonic brain tissues separately,and analyzed by high performance thin layer chromatography(HPTLC).RESULTS CDH and CTH in glioma,while CMH and CTH in embryonic brain tissues,showed an increased expression as compared with adult brain.Both of the glioma and embryonic brain tissue had no expression of globoside.CONCLUSIONS Alteration of the N GSLs structure is associated with oncogenic transformation of the cells.One of the characteristic changes of N GSLs is oncofetal change.
