摘要
目的探讨细胞共同抗原CD45RB在缺血性脑损伤中作用。方法选用SD成年大鼠阻塞大脑中动脉2h,再灌流0.5~48h制成短暂局灶性脑缺血,用CD45RB对缺血性脑组织进行免疫组化染色。结果正常组及假性手术组的脑组织有散在的神经元弱表达CD45SRB。缺血脑组织,皮质层锥体细胞、纹状体及下丘脑区的神经元CD45RB强阳性,其胞核形态及对苏木素着色均正常;轻度变性的神经元弱表达CD45RB:细胞核固缩死亡的神经元CD45RB阴性。结论本实验证实CD45RB在缺血时形态正常的神经元呈强表达,其重要意义尚需进一步研究。
Objective To observe the role of CD45RB in transient focal ischemia in rat. MethodsThe focal ischemia was made by occluing the middle cerebral artery for 2 hours and reperfusion 0. 5-48hours. The ischemie brain tissue was studied by monoclonal antibody against CD45RB. Results Thescattered neurons weakly expressed CD45RB in normal and sham-operated animals. Following transientfocal cerebral ischemia,CD45RB was strongly induced in larger pyramidal neurons of cortical as well asthe bigger neurons of striatum. hypothalamus. The nuclear shape and hematoxylin stainning of CD45 positive neurons were normal: the slightly degenerative neurons weakly expressed CDRB; the dead neuronsdidn't expressed CD45RB. Conclusion The data conformed that the normal neurons based on morphology strongly expressed CD45RB. The significance of CD45 expression in neurons after transient focal cerebral ischemia needs to be investigated.
