摘要 目的:研究肾上腺髓质素(AM)对内皮素1(ET-1)、血管紧张素(Ang)促肺动脉平滑肌细胞增殖的影响。方法:体外培养兔肺动脉平滑肌细胞,采用3H-TdR掺入法观察不同剂量AM对ET-1,Ang促肺动脉平滑肌细胞蛋白质合成的影响。结果:1×10-9mol/L的ET-1,1×10-8mol/L的Ang明显增加培养的平滑肌细胞3H-TdR掺入量。1×10-7~2×10-8mol/L浓度的AM明显抑制1×10-9mol/L的ET-1引起的平滑肌细胞3H-TdR掺入量的增加;1×10-7~5×10-8mol/L浓度的AM明显抑制1×10-8mol/L的Ang引起的平滑肌细胞3H-TdR掺入量的增加。结论:AM能剂量依赖地抑制ET-1,Ang促肺动脉平滑肌增生,可能在肺血管病理变化中起一定作用。 Objective: To study effects of adrenomedullin(AM) on proliferation of pulmonary artery smooth muscular cells(PASMC) induced by endothelin 1 or angiotensin Ⅱ. Methods: The levels of trichloroacetic acid insoluble tritiated thymidine( 3H TdR) of cultured rabbit PASMC was measured in medium that contained endothelin 1(ET 1) or angiotensin Ⅱ(Ang Ⅱ) or with different concentrations of AM. Results: The level of 3H TdR significantly increased in PASMC cultured with 1×10 -9 mol/L ET 1 and 1×10 -8 mol/L Ang Ⅱ, which could be inhibited by 1×10 -7 ~5×10 -8 mol/L AM in concentration dependent manner. Conclusion: The results suggest that AM can significantly inhibit the proliferation of PASMC stimulated by ET 1 or AngⅡ and may play a role in the pulmonary physiological changes.
