摘要
目的:建立既适用于常规病理检验又可进行免疫组化及核酸原位杂交研究的骨髓活组织检查方法。方法:活检标本10例,用Low yFMA液固定、脱钙,石蜡包埋。切片行常规染色、TGF-β1 和MIP-1α免疫组化及地高辛标记DNA探针m RNA原位杂交。结果:HE染色、鼠源性单抗和兔源性多抗的免疫组化及不同长度探针的m RNA原位杂交均取得理想的效果。结论:LowyFMA液固定、脱钙后石蜡包埋的骨髓活检组织不仅能满足常规病理检验需要,且适用于不同条件的免疫组化及原位杂交研究。
Objective: To establish a m ethod that may fulfill the need for routine pathological exam ination, im munohistochem istry and in situ hybridization researches ofbone m arrow biopsy sam ples. Methods: After fixation and decalcification with Lowy FMA solution, bone m arrow biopsy sam ples were stained routinely as w ellas w ith imm unohistochem istry and in situ hybridization of TGF-β1 and MIP-1α. Results: The sam ples show ed a good preservation of m orphology in routine examination while im munohistochem istry and in situ hybridization with differentantibodiesand probesachieved perfectstaining too. Conclusion: Fixation and decalcification ofbonem arrow biopsy sam plesw ith Low y FAM solution m ay notonly beused as a routine method for clinicalbone m arrow pathologicalexamination, butalso provide a perfectprocedure for im m unohistochem istry and in situ hybridization research.
