摘要
目的:建立一人脑胶质瘤细胞系并探讨其生物学特性。方法:取胶质瘤新鲜手术标本,经组织块法培养并克隆、建株,采用光镜、电镜、免疫组化、流式细胞术、染色体分析及异种移植瘤实验对细胞株生物学特性进行观察。结果:原位肿瘤、细胞株及移植瘤标本经电镜证实为星形细胞源性,免疫组化呈GFAP、Vim entin及VEGF阳性,异种移植成瘤率高。与SHG-44细胞比较,GFAP表达强,Vim entin 表达弱。结论:CHG-5为一恶性胶质瘤细胞系,且其生物学特性可能与SHG-44细胞有所不同。
Objective: To establish a hum an m alignant human gliom a cell line and investigate its biological features. Methods: Surgically resected specim en of a case of m ixed astrooligodendrocytom a of grade Ⅱwas collected. After prim ary culture and cloning, a cellline designated asCHG-5 was established and its biologicalfeatures w ere studied with opticaland electron m icrosopies, im m unocytochem istry, flow cytom e try, karyotype analysis and experim ental transplantation to nude m ice.Results: The surgical specim en, cellline and xenografted tumorexhibited thecharacteristics ofgliom a underelectron m icroscopy. Im m unohistochemical staining for GFAP, vimentin and VEGF were positive. There was a high rate of xenografted tum orform ation in nude m ice. CHG-5 cellline showed a strongerreaction to GFAPand weaker reaction to vim entin than SHG-44 cellline.Conclusion: CHG-5 cellline is cellline ofm alignant gliom a and certain biologicalcharacteristicsofCHG-5 are differentfrom those ofSHG-44.
