摘要
目的探讨中国常州地区汉族人群急性冠脉综合征(ACS)患者血清单核细胞趋化蛋白-1(MCP-1)水平与MCP-1基因-2518G/A多态性和相关因素的相关性。方法对101例ACS患者(ACS组)和73例因胸闷(痛)住院经冠脉造影检查证实无冠脉病变者(对照组)用聚合酶链式反应限制性酶切片段长度多态性技术检测了MCP-1基因-2518G/A多态性,用ELISA方法检测了血清MCP-1水平。结果 MCP-1基因-2518 G/A位点的AA、GG和GA基因型及等位基因频率在ACS组和对照组间的差异均无统计学意义(均P>0.05);血清MCP-1水平(M/IQR)ACS组显著高于对照组(263.65/385.78 pg/ml vs 100.71/134.02 pg/ml,P<0.01);在ACS组和对照组的组内任何基因型之间,血清MCP-1水平(M/IQR)的差异均无统计学意义(均P>0.05);ACS组血清MCP-1水平(M/IQR)与年龄、糖尿病和吸烟成正相关,与男性成负相关,与高血压和高血脂无关。结论在中国常州地区汉族人群中,ACS和非冠心病患者的血清MCP-1水平与MCP-1基因-2518G/A多态性无关联,与某些冠心病易患因素相关。
Objective To investigate the association of serum monocyte chemoattractant protein-1(MCP-1) level with polymorphism in the MCP-1 gene-2518 G/A and the relevant factors in patients with acute coronary syndrome(ACS) in Chinese Han population of Changzhou region.Methods The polymorphism in the MCP-1 gene-2518 G/A was genotyped by PCR-RFLP and the serum MCP-1 level was measured by ELISA in 101 patients with ACS(ACS group) and 73 subjects(control group) with chest pain who were free from coronary heart disease tested by coronarography.Results No significant differences in the frequencies of the AA,GG and GA genotype and allele frequency of MCP-1 gene-2518 G/A locus were found between patients with ACS and subjects freed from coronary heart disease(all P0.05).Serum MCP-1 level(M/IQR)in ACS group was significantly higher than that in the control group(263.65/385.78 pg/ml vs 100.71/134.02 pg/ml,P0.01).No significant difference in serum MCP-1 level(M/IQR) was found among the any of the genotypes of MCP-1 gene-2518G/A in ACS group and the control group(all P0.05).Serum MCP-1 level(M/IQR) was positively correlated with the age,diabetes and smoking,negatively correlated with the male,and unrelated with the hepertention and hyperlipidemia in ACS group.Conclusions Serum MCP-1 level in patients with ACS and subjects freed from coronary heart disease is irrelevant with the MCP-1 gene-2518G/A polymorphism and related to some risk factors of coronary heart disease in Chinese Han population of Changzhou region.
