摘要
目的:建立大鼠跟腱周围炎病理模型,揭示针刺防治肌腱周围炎的作用机制。方法:24只Wistar大鼠随机分为对照组、造模组和针刺干预组,采用"跳跃运动"方法制作大鼠跟腱周围炎的动物模型,6周后取材进行HE染色,免疫组织化学法检测HIF-1α和VEGF的表达。结果:(1)模型组大鼠跟腱及其周围组织出现明显的炎性反应病理特征,腱组织、肌肉以及软骨组织的HIF-1α和VEGF的表达水平均明显高于对照组(P<0.01)。(2)针刺干预组的腱组织和肌肉中HIF-1α和VEGF表达水平高于对照组(P<0.05或P<0.01),软骨组织则均无明显差异,但针刺干预组与模型组比较,腱组织、肌肉以及软骨组织的HIF-1α和VEGF的表达水平明显下降(P<0.05或P<0.01)。结论:"跳跃运动"是建立跟腱周围炎动物模型的可行方法,改善组织内部缺氧环境可能是针刺防治肌腱周围炎重要途径之一。
Objective:To explore the control mechanism of acupuncture revealed paratendinitis,a rat pathological model of paratendinitis was developed.Methods:24 Wistar rats were randomly divided into control group,model group and the acupuncture treatment group.A"jumping movement"approach was used to make a rat model of paratendinitis.Six weeks after operation,rat tissue were stained by routine HE.HIF-1α and VEGF expression were detected by Immunohistochemical method.Results:(1)rat tendon and surrounding tissue showed obvious pathological features in Model group.HIF-1α and VEGF expression in Tendon,muscle and cartilage tissue in Model group were significantly higher than in control group(P0.01).(2)HIF-1α and VEGF expression of tendon and muscle in Acupuncture intervention group were higher than in control group(P0.05 or P0.01).There was no significant difference in cartilaginous tissue.But HIF-1α and VEGF expression levels of tendon,muscle and cartilage tissue in acupuncture intervention group were significantly decreased,compared with that of in model group(P0.05 or P0.01).Conclusion:"Jumping movement"is a feasible way to built a rat pathological model of paratendinitis.Improving Hypoxic environment within the organization may be one of important ways that acupuncture revealed paratendinitis.
