摘要
目的:建立水蛭仿生酶解有效部位HPLC指纹图谱的鉴定标准。方法:采用依利特SinoChrom ODS-BP色谱柱;0~25min,甲醇(1%~10%)~水梯度洗脱;柱温35℃;流速0.8 mL.min-1;检测波长254nm。结果:在相同色谱条件下获得6批水蛭仿生酶解有效部位的指纹色谱图,其中8个色谱峰分离较好,均达到指纹图谱的技术要求。结论:HPLC指纹图谱中8个共有峰的相对保留时间比值(RRT)和相对峰面积比值(RPA)可用于水蛭仿生酶解有效部位的质量控制。
Objective:To establish the HPLC fingerprint detecting standard of small bionic zymolysis leeches peptides by reversed phase high performance liquid chromatography.Methods:The chromatographic conditions were as follows:a SinoChrom-ODS-BP column was used;the mobile phases methanol(1%~10%)-water from 0~25 min with gradient elution;column temperature 35℃;the flow rate 0.8 mL.mim-1 and the UV absorbance detection at 254 nm.Results:Under the same selected chromatographic conditions,the HPLC fingerprints of 6 batch small bionic zymolysis leeches peptides were obtained and the separation and correlation of 8 common peaks are good according to the technical requirements of fingerprint.Conclusion:The retention times and peak area ratios on HPLC fingerprint of 8 common peaks can be used as the important criterion.
