虾夷扇贝生殖腺多肽的制备及分离 被引量:13

摘要 为了对扇贝加工副产物进行高值化利用,采用中性蛋白酶,分别以变性(100℃,10min)和未变性虾夷扇贝生殖腺匀浆为底物,经50℃酶解3h后制备不同的生殖腺水解液。结果表明,中性蛋白酶水解雌性虾夷扇贝生殖腺的效果优于雄性生殖腺,并且虾夷扇贝生殖腺经变性处理后,水解度和肽得率得到显著提高。与未变性处理组雌性虾夷扇贝生殖腺相比,变性处理组的水解度和肽得率分别提高了36.16%和94.78%。雌性虾夷扇贝生殖腺水解液经Sephadex G-25凝胶层析分离,发现变性处理后雌生殖腺水解液的组分更为集中,且具有一定的还原能力。经Superdex Peptide 10/300GL凝胶过滤色谱分析,变性处理后的雌生殖腺水解液分子质量主要分布在0.25~5ku,占水解液的74.78%。因此,利用中性蛋白酶并结合变性预处理,对雌性生殖腺可以进行有效水解,从水解液中可分离出具有还原能力的多肽。 Both raw and heat denatured gonad homogenate from Patinopecten yessoensis were hydrolyzed with neutral proteinase at 50 ℃ for 3 h to prepare peptides.The hydrolytic efficiency of female scallop gonads is higher than that of male scallop gonads.The hydrolytic degree and yield of peptides were increased by 36.16% and 94.78% respectively when the denatured female scallop gonads were used.The narrow band can be obtained when the female denatured gonad hydrolysates were separated by Sephadex G-25 gel filtration chromatography,which has some reducing power.The molecular weight distribution of 74.78% peptides in female denatured gonad hydrolysates was ranged from 0.25 to 5 ku.The results suggested that the peptides with reducing power can be prepared by hydrolysis of female denatured gonad with neutral proteinase.
