摘要
目的比较RNA和马洛替酯对肝组织超微结构的影响。方法72只家兔随机均分为正常组、病理组、RNA组和马洛替酯组。于用RNA或马洛智酯后第40、70、100天,每组随机选6只取肝和血,分别通过透射电镜观察肝组织超微结构变化,用紫外分光光度计测定肝组织羟脯氨酸(HYP),用RIA法测定血浆透明质酸(HA)含量。结果RNA组和马洛替酯组贮脂细胞、Kupffer细胞增生活跃程度均明显低于病理组,贮脂细胞周围胶原纤维明显减少,肝细胞线粒体粗面内质网增多且排列整齐,粗面内质网上附着颗粒增多;RNA组和马洛替酯组于100天时血浆HA(分别为4.66μg/ml±1.35μg/ml,6.35μg/ml±2.44μg/ml)和肝组织HYP(7.24μg/mg±1.24μg/mg,7.21μg/mg±1.39μg/mg肝粉)均显著低于病理组相同阶段的HA(10.16μg/ml±2. 52μg/ml,P<0.01)和HYP(10.85μg/mg±1.73μg/mg,P<0.01)。结论提示RNA和马洛替酯均能改善肝组织细胞超微结构,减少贮脂细胞胶原纤维的生成量,阻抑肝纤维化的进一步发展。
Objective To compare the effect on the ultramicrostructure of rabbit's liver with schistosomiasis liver fibrosis after treatment with RNA and malotilate. Methods Seventy-two do mestic rabbits were randomly and equally divided into normal group, pathologic group, RNA group, and malotilate group. On the 40th, 70th, 100th day after RNA or malotilate administered,6 rabbits taken randomly from each group in the above 4 groups were killed and the hepatic tissue ultramicrostructure was observed under electronic microscope, the level of hydroxyproline (HYP ) in liver tissue was tested with violet-spectrophotometer and hyaluronic acid (HA) in plasma was de tected with RIA. Results In RNA group and malotilate group the proliferated degree of fat-storing cell and kupffer cell were obviously less than that of the pathologic group, collagenic fiber around fat-storing cell apparently decreased, the hepatocellular cristae, rough endoplasmic reticulum (RER) and granule on the RER were increased. RER was regularly put in order. On the 100th day the levels of HA (4. 66μg/ml ±1. 35μg/ml in RNA group, 6. 35μg/ml ± 2. 44μg/ml in malotilate group ) in plasma and HYP (7. 24μg/mg ± 1. 24μg/mg liver powder in RNA group, 7. 21 μg/mg± 1. 39μg/mg in malotiate group) in liver were distinctly lower than those of HA (10. 16μg/ml ± 2. 52μg/ml and HYP (10. 85μg/mg ±1. 73μg/mg) of pathologic group in the same stage (all P< 0. 01). Conclusion RNA and malotilate might improve the ultramicrostructure of hepatocellular, decrease the amount of collagenic fiber produced by fat-storing cell, prohibit further development of liver fibrosis.
