塑料条快速ELISA诊断日本血吸虫病的影响因素分析和操作规范化研究

STUDY ON ANALYZING THE AFFECTION FACTORS AND DEVELOPING THE OPTIMAL OPERATION CONDITIONS OF THE FAST PLASTIC-STRIP ELISA FOR DIAGNOSIS OF SCHISTOSOMIASIS JAPONICA

目的对塑料条快速ELISA诊断日本血吸虫病的影响因素进行分析，并建立规范化的操作方法。方法比较抗原、待检血清、酶结合物和底物等因素对检测结果的影响，选择最佳操作流程，并与塑料条ELISA、常规ELISA方法进行比较。结果选择了抗原、待检血清、酶结合物和底物的最佳实验条件，确立了规范化的塑料条快速ELISA操作流程，并与塑料条ELISA，常规ELISA进行了比较。3种检测方法的敏感性分别为95．0％、96．0％、95．0％，特异性均为99．0％。结论 塑料条快速ELISA方法具有与常规方法相近似的敏感性和特异性，且方法简捷，适于现场应用。

Objectives To analyze the affection factors of lhe fast plastic-strip ELISA (FPS ELISA ), and to develop and optimize the operation procedure. Methods The optimal operational procedure was selected by comparing the reaction conditions of the antigen, serum, enzyme conju gate and substrate. In adition, some sera were tested by comparing with plastic-strip ELISA (PS ELISA ) and conventional ELISA. Results The optimal reaction conditions of antigen, serum, conjugate and substrate were selected, and the optimal operation procedure of FPS-ELISA was es tablished. The sensitivities of FPS-ELISA. PS-ELISA and conventional ELISA were 95. 0%,96. 0% and 95. 0% respectively. The specificties of three assays were the same as 99. 0%. Conclu sion The FPS-ELISA developed has kept the advantage of sensitivity and specificity of the conven tional ELISA, also has the advantages of simple, easy to use. It is suitable for applying in the field.
