Detection of Minimal Residual Disease in Remission Patients with Acute Promyelocytic Leukemia by Reverse Transcriptase polymerase Chain Reaction

摘要 用逆转录 -聚合酶链反应 ( RT- PCR)技术 ,检测了 2 2例早幼粒细胞白血病 ( APL )缓解患者 PML- RARα融合基因。结果显示阳性率在缓解时间 1年内 ( 11/14 )较 1~ 2年 ( 2 /5 )及 3年以上( 0 /3)明显高 ,持续阳性多预示着复发。认为检测 PML- RARα融合基因 ,可作为监测 APL病情转归、评价疗效的重要指标。 Objective:To investigate the correlation between minimal residual disease (MRD) and relapse in postremission patients with acute promyelocytic leukemia (APL) during the different periods.Methods:The PML RAR α fusion gene were detected by using reverse transcriptase polymerase chain reaction(RT PCR)in 22 remission patients with APL.This assay has higher sensitivity of 10 -5 .Results:The positive rate of the remission patients within 1 year (11/14)was significantly higher than that of 1 to 2 years (2/5) and more than 3 years (0/3),persisitently positive result closely associated with the relapse.Conclusion:Detection of PML RAR α fusion gene was a valuable parameter in monitoring the prognosis of APL and evaluating the therapeutic effectiveness.
