摘要 目的探讨高氧对体外培养的人胚肺成纤维细胞(HELF)增殖、凋亡的影响。方法将生长良好的HELF传代后,用无血清培养基培养24 h,分别置于空气及90%高氧的细胞培养箱中,培养5 d,绘制细胞生长曲线,免疫组化法检测增殖细胞核抗原(PCNA)蛋白表达,流式细胞术检测细胞凋亡率。结果与空气相比,高氧抑制HELF的增殖,诱导HELF凋亡(P<0.05或<0.01),且随高氧暴露时间延长,呈现明显的时间依赖关系。结论体外高氧对HELF具有明显的抑制细胞增殖和促进细胞凋亡作用。 Objective To develop the direct effect of hyperoxia on proliferation and apoptosis in human embryo lung fibroblasts in vitro.Methods After being passaged,HELFs were cultured for 24 hours in DMEM with no fetal bovine serum,then randomly divided into air and 90% O2 group.Cells were cultured for 5 days.Cellular growth curve was drew,the expressions of PCNA protein in HELF were determined by immunohistochemistry,the apoptosis of cells were analyzed by flow cytometry.Results As compared with air group,90% O2 caused a decreased cell proliferation and an increased cell apoptosis of HELF.Conclusion Hyperoxia could result in growth arrest and cell apoptosis in vitro.
