摘要
目的探讨重症急性胰腺炎(SAP)并肾损害大鼠肾组织Livin、Caspase-9蛋白变化及意义。方法将90只大鼠随机分为假手术组(A组)、SAP并肾损害组(SAP组)、褪黑素(MT)干预组(MT组)各30只,观察其胰腺、肾组织病理改变,检测血清淀粉酶(AMY)、肌酐(Cr)、丙二醛(MDA)、超氧化物歧化酶(SOD)活性及肾组织Livin、Caspase-9表达;分析SAP并肾损害与Livin、Caspase-9表达的相关性。结果光镜下,A组胰腺、肾组织均正常;SAP组随造模时间延长病变逐渐加重;MT组病理改变较SAP组减轻。与A组比较,SAP组AMY、Cr、MDA、Caspase-9升高,SOD、Livin降低(P均<0.01);与SAP组比较,MT组AMY、Cr、MDA、Caspase-9降低,SOD、Livin升高(P均<0.05)。SAP组SOD与Cr、Caspase-9表达呈负相关,MDA与Cr、Caspase-9表达呈正相关(P均<0.05);MDA、SOD与Livin表达无相关性(P均>0.05)。结论 Livin、Caspase-9蛋白变化可能参与SAP大鼠的肾损害,外源性MT对SAP并肾损害有保护作用。
Objective To investigate the significant changes of Livin,Caspase-9 protein in rats of severe acute pancreatitis(SAP) associated with renal injury.Methods Ninety SD rats were randomly divided into three groups: the sham operation group(group A),SAP associated with renal injury group(group SAP) and melatonin intervention group(group MT),whose pathologic changes of pancreas and kidney were observed.Furthermore,the levels of serum amylase(AMY),creatinine(Cr),malondialdehyde(MDA),superoxide dismutase(SOD) and the expressions of Livin and Caspase-9 in renal tissue were detected.Finally,the relevance between renal injury in SAP and Livin and Caspase-9 was analyzed.Results Pancreas and kidney tissue in group A were observed by light microscopy,and showed no evident lesion;the pathological lesion in group SAP was gradually aggravated with extension of molding time;the pathological lesion in group MT at each time point was more remissive than that in group SAP.Compared with the group A at each time point,expression levels of the serum AMY,Cr,MDA and Caspase-9 in group SAP were increased(P0.01),and which were decreased in the SOD and Livin(P0.01);and the results were identical when compared to the group MT(P0.05).In group SAP,the expression levels of Caspase-9 and Cr were negatively correlated with SOD(P0.05),and which were positively correlated with MDA(P0.05);the MDA and SOD had no relevance to Livin respectively(P0.05).Conclusion The changes of Livin,Caspase-9 protein may be associated with the severity of renal injury of SAP rats,and exogenous melatonin may have a protective effect on renal injury in SAP.
