摘要
目的:为寻找一种简便、经济、灵敏度高而不需使用放射性同位素的生物活性检测方法对成纤维细胞生长因子(FGF)进行检测。方法:利用肝素对Balbc3T3 细胞株进行诱导,然后运用结晶紫比色法对其进行生物活性检测。结果:最大吸光度A值的一半为0 .315,所对应的aFGF浓度为24 pgml,而没有肝素诱导的最大A值一半为0.16 ,所对应的浓度为156pgml。结论:肝素的存在提高了结晶紫法的灵敏度。因此是一种简便、快捷而值得推广应用的FGF生物活性检测法。
Objective:Fibroblastgrowthfactor(FGF)isoneoftheimportantcytokinesfor wound healing.Itcanacceleratetheregenera tion ofblood vessel,nerve andepidermis.Tolookfora bioassayofFGFthatisnotonlyeasy,economicand highsensitivity ,butalsonotinvolv ingradioisotopesuch as 3 H TdR.Methods:The authors used heparinto inducethe Balbc 3T3 cells,and then the crystal violet colorimetric analysis(CVCA) was used to measureits boiogicalactivity .Results:Itshowedthatthe half maximalof Avalue was stimulated by 24 pgml aFGFwithinducing by heparin,whilethat wasstimulated by 156 pgmlaFGFwithout heparin inducing .Conclusion :The existing ofheparin could enhancethesensitivity of CVCA.It may be an easy,sensitive and valuable methodto popularizefor FGFbioassay.
