摘要
葡萄糖淀粉酶(糖化酶)和异淀粉酶是重要的工业酶制荆,糖化酶属于糖苷水解酶,它将淀粉及类似物水解成D(+)-葡萄糖;异淀粉酶属于脱支酶.近年来,对于葡萄糖淀粉酶固定化的研究较多。然而对于异淀粉酶固定化的研究甚少,对于葡萄糖淀粉酶和异淀粉酶的共固定研究没有发现.我们以纤维素为载体,通过重氮化法固定了葡萄糖淀粉酶和异淀粉酶,通过L9(3^4)的正交试验对固定化条件参数进行了优化.并对固定化酶的性质进行了研究,该固定化酶在pH4.6、40℃,以10%的可溶性淀粉为底物进行了分批试验,测得操作丰衰期为12.29h,米氏常数Km为7.18g/L,糖化试验的DE值为92%.
Glucoamylase is a kind of glycoside hydrolase's. Glucoamylase is due to the abili- ty of hydrolyzing starch or the similarity of starch into D (+)-glucose. Iso-amylase is due to debauching. They play an important role in industrial production. A great study has been made until now on immobilization of Glucoamylase, but a few on immobilizations of iso-amy- lase, few studies on immobilization of Glucoamylase and iso-amylase. Glucoamylase and isoamylase were simultaneously immobilized on cellulose by using diazole method. By l.o (34) orthogonal experiment, the optimal conditions for co immobilization were established, and the properties of the co immobilized enzymes were studied. The enzyme system could directly converted 10 % starch hydrolyses to glucose under the conditions of pH=4.6, 40℃.The results show that the working half-life of immobilized mixed enzyme was 12.29 h. The Km was about 7.18 g/L.The DE was about 92 %.
